Description Applied StemCell’s total RNA passes an extensive testing to ensure a high degree of purity and intactness. RNA is isolated using a modified guanidinium thiocyanate method.1 Residual genomic DNA is removed with RNAase-free DNAase I. Applied StemCell’s total RNA can be used in downstream applications such as RT-PCR and real-time RT-PCR; differential display; cDNA synthesis; Northern, dot, and slot blot analyses; primer extension; poly A+ RNA selection; RNAase/S1 nuclease protection; and microarrays.

Test Conditions RNA quality is determined visually by analyzing 18S and 28S ribosomal bands using non-denaturing agarose gel electrophoresis. The RNA purity level is determined by spectrophotometry to obtain the A260/A280 ratio, which must range from 1.8-2.1. Plus, the RNA functionality is assayed by RT-PCR using primers for housekeeping gene GAPDH. RNA is confirmed to be DNA-free by RT-PCR.
Storage and Handling
Handling Instructions RNA products are sensitive to degradation by temperature changes or exogenous ribonucleases that result during the handling processes. Must wear gloves at all times when handling RNA. All reagents used, tubes, and barrier pipettes must be RNAase-free.
For thawing RNA: place tube in ice and thaw to completion. Vortex tube for a few seconds and spin down RNA quickly and place in ice once again. To minimize freeze-thaw cycles (< 3), aliquot RNA as desired.
Storage Conditions Store total RNA at -80°C. Can be stored at -20°C for short term in a frost-free freezer. Avoid multiple freeze-thaw cycles. To prevent any possible RNAase contamination, always wear gloves when handling RNA. For maximum recovery of product, centrifuge the original vial after thawing and before removing the cap.
