Overview

Microglia are the primary immune cells in the central nervous system (CNS) and play a crucial role in: normal brain development and neuronal function by maintaining neuronal homeostasis and synaptic plasticity; inflammatory response and pathogenesis of several neurological disorders such as Alzheimer’s disease and Parkinson’s disease. However, primary microglial cells are difficult to source especially primary, normal microglia from healthy patients. To overcome the sourcing problem associated with primary microglia, we developed an efficient method to derive high-quality and consistent microglia cells from human iPSCs which recapitulate the phenotype and functional parameters of primary microglia and in vivo microglial cells. Our erythromyeloid induction protocol mimics in vivo activation pathway for development of microglia in brain from hematopoietic stem cells.

  • Delivered as robust, mature cells with morphology of human primary microglia
  • iPSC derived using different reprogramming strategies from fibroblast/ PBMC/ CD34+ cord blood cells
  • From healthy/ disease/ engineered iPSCs
  • Well-characterized “Master” control, human iPSC lines available for a ready start to your differentiation projects as well as control lines for your disease modeling experiments
  • High purity (>90%) TMEM119, and P2Ry12+/P2Y12+ microglial-specific markers
  • Optional! RT-PCR, Western Blot, proliferation assays, phagocytosis, functional assay

Applications:

  • Neurodevelopment and neuroinflammation research
  • Disease modeling for neuroinflammation and neurodegenerative diseases
  • Mimic in vivo environment by co-culture with iPSC-derived neurons and glial cells for cell-cell interaction studies and signaling pathway
  • In vitro and in vivo (adoptive transfer) cell line model to investigate microglial dysfunction in CNS disorders
  • High throughput drug target discovery, efficacy and toxicity testing

Timeline = 12-14 weeks
