CRISPR Service - Guide RNA Vector Evaluation (Validation)
Applied StemCell now provides a new service for evaluating the CRISPR/Cas9-gRNA system. We use two methods to perform functional tests:
1) Cell Based Assay: using mouse N2A cells or human HEK293 cells to validate the gRNAs. The assay is based on the generation of PCR products that are subsequently hybridized to mismatches in heteroduplexed DNA, which is then treated and cleaved by a nuclease.
2) Cell-Free System: based on a sequence-specific cleavage assay of episomal plasmids. The efficiency of cleavage is detected by PCR.
You send us your plasmids (gRNAs and CAS9 in one vector or separate vectors, 3 ug each), and you will receive a report including high resolution PCR gel images in 3-4 weeks to help you determine which gRNA is the best to use.
