Case Study 1:

  • Test if iNK cells (ASE-9708) are functional against hematopoietic tumor cell line, K562

Figure 1: Natural Killer (NK) Cell Cytotoxicity Assay Fluorescence Analysis. The ASE-9708 iPSC-derived Natural Killer Cells successfully killed the K562 target cells (Green). (Images captured at 2 hours, 3 hours, and 16 hours; Top Row: Control; Middle Row: 10:1 ratio of NK Cells to K562 Cells; Bottom Row: 5:1 ratio of NK Cells to K562 Cells)

Case Study 2:

  • Test if iNK cells (ASE-9708) are functional against hematopoietic tumor cell line, K562
  • Compare iNK cell function to primary NK cells obtained from PBMC

Figure 2. Comparison of iNK cell (ASE-9708) and primary NK cell (from PBMCs) function against hematopoietic tumor cell line, K562. The K562 cells were stained with Calcein AM for 30 minutes. The labeled K562 cells were mixed with NK cells at different E/T ratios (Effector: NK, Target: labeled K562). Fluorescence was read by microplate reader at wavelength excitation 490nm, emission 520nm. Standard curve was generated by measuring different numbers of labeled K562 cells only. The images were taken under fluorescence microscope.

Figure 3. K562 cell lysis percentages for different E/T ratios (Effector: NK, Target: K562) captured at 3 time points. K562 cell lysis percentages were documented after 1 hour, 2 hours, and 16 hours.

Figure 4. Comparision of the resulting K562 cell lysis percentages for different E/T ratios (Effector: NK, Target: K562) captured at distinct time points. K562 cell lysis percentages were recorded after 1 hour, 2 hours, and 16 hours.
