The episomal kit for human induced pluripoten stem cell (iPSC) generation is a cost-effective combination of multiple episomal vectors which encode the essential reprogramming factors, Oct-3/4, Sox2, Klf4, and l-Myc. Optimized by the ASC team, our episomal mix has been experimentally proven to induce pluripotency for a number of different somatic cell types. The episomal reprogramming kit is one of the best choices for producing footprint-free iPSCs, providing a source of iPSCs for all stages of your pluripotent stem cell research. Unlike other reprogramming methods, such as lentivirus and retrovirus, this kit is able to efficiently generate transgene-free, virus-free and footprint-free iPSCs in both feeder and feeder-free conditions.
The episomal vectors are introduced into the cell by electroporation and the reprogramming efficiency is enhanced by puromycin selection. The reprogramming protocol is quite simple yet highly effective with efficiencies in the range of 0.05-1.0% depending upin the cell type being reprogrammed. Reprogramming enhancers, such as PD0325901, CHIR99021, A-83-01, hLIF, and HA-100, are not required for reprogramming.
