Humanized PBMC and HSC CD34+ Mouse Models

Human peripheral blood mononuclear cell-engrafted M-NSG mice are perfect models for short-term experiments requiring mature T cells, including immuno-oncology, and GVHD research.

Our humanized models are generated using our M-NSG  mice through the injection of healthy human donor peripheral blood mononuclear cells (PBMCs) or CD34+ hematopoietic stem cells (HSCs). After a few weeks of reconstruction following the injection of PBMCs or CD34+ HSCs, the mice develop a human-like immune system and are ready to be shipped.

1. Ficoll density gradient centrifugation is used to purify human peripheral blood mononuclear cells (PBMCs) from cord blood

1. 4-weeks-old M-NSG female mice are irradiated with 1Gy 12h

2. The 5E6 Human PBMCs are injected into the M-NSG mice

2. 1E5 human CD34+ hematopoietic stem cells derived from umbilical cord blood are injected into the mice

3. After 3 weeks of reconstruction, a relatively well-developed human-derived immune system develops

3. 16 weeks after the transplantation of human CD34+ HSC (hematopoietic stem cells) into the M-NSG mice, the human HSCs develop a relatively well-developed human-derived immune system
